Pinostrobin is a natural compound that has been shown to be effective in the treatment of type 2 diabetes. It has minimal toxicity, with no significant cytotoxicity observed in vitro at doses up to 10 μM. Pinostrobin inhibits mitochondrial membrane potential and impairs mitochondrial functions, leading to decreased glucose uptake and increased insulin sensitivity. The hypoglycemic effect of pinostrobin was also demonstrated in vivo using an animal model. The mechanism of action is believed to be due to inhibition of various enzymes involved in the production of reactive oxygen species and reactive nitrogen species by mitochondria, including caffeic acid-O-methyltransferase (COMT). Pinostrobin has been shown to inhibit the activity of COMT through competitive inhibition, which may also account for its lack of cytotoxicity.
