MZ 1 is a thuringiensis derivative that inhibits the activity of protein kinases. The inhibitory activity of MZ 1 is due to hydrogen bond formation with the active site serine residue and competitive inhibition of ATP binding. In vitro, MZ 1 has been shown to be cytotoxic against prostate cancer cells and skin cancer cells, as well as inhibiting the growth of cultured human breast cancer cells. It may also have anti-inflammatory properties due to its ability to suppress transcriptional regulation by all-trans retinoic acid (RA). MZ 1 is a molecule in clinical development for the treatment of HER2+ breast cancers.
