Crkl is a cell surface protein that is expressed in the cytoplasm of cells and on the cell membrane. It has been shown to be involved in signal transduction, apoptosis, and cancer progression. Crkl has also been shown to regulate gene transcription and tyrosine kinase activity. Crkl is found at high levels in solid tumours such as breast, prostate, lung, colon, stomach, pancreas, brain and ovarian cancers. Expression of Crkl is important for epithelial mesenchymal transition (EMT), which plays a role in metastasis. Treatment with dasatinib or monoclonal antibodies targeting the tyrosine kinase domain of Crkl may be an effective way to inhibit tumorigenesis. Dasatinib also inhibits growth factor production by stromal tumour cells. In addition to its effects on cancer cells, Crkl also regulates epidermal growth factor (EGF) production by keratinocytes and stem cell factor
