The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human CASP9. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human CASP9. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human CASP9, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human CASP9 in the samples is then determined by comparing the OD of the samples to the standard curve.Color and Shape:Colourless TransparentliquidRat CASP9(Caspase 9) ELISA Kit

Caspase-9 is an initiator caspase, encoded by the CASP9 gene.Binding of caspase-9 to Apaf-1 leads to activation of the protease which then cleaves and activates caspase-3. Proteolytically cleaves poly(ADP-ribose) polymerase (PARP).Cleaved-Caspase-9 p35 (D315) rabbit pAb
CASP9 encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. Caspase 9 can undergo autoproteolytic processing and activation by the apoptosome, a protein complex of cytochrome c and the apoptotic peptidase activating factor 1; this step is thought to be one of the earliest in the caspase activation cascade. Caspase 9 is thought to play a central role in apoptosis and to be a tumor suppressor. Alternative splicing results in multiple transcript variants.Caspase-9 (phospho Thr125) rabbit pAb
