Leflunomide

Description:
Leflunomide is an immunomodulatory drug primarily used in the treatment of rheumatoid arthritis and psoriatic arthritis. It functions as a prodrug, which means it is metabolized in the body to its active form, A77 1726, that inhibits the enzyme dihydroorotate dehydrogenase. This inhibition disrupts the de novo synthesis of pyrimidines, leading to a reduction in lymphocyte proliferation and modulation of immune responses. Leflunomide is characterized by its relatively long half-life, allowing for once-daily dosing. It is typically administered orally and is known for its potential side effects, which can include liver toxicity, gastrointestinal disturbances, and effects on blood cell counts. The drug is contraindicated in pregnancy due to its teratogenic effects. Additionally, leflunomide is often used in combination with other disease-modifying antirheumatic drugs (DMARDs) to enhance therapeutic efficacy. Its chemical structure includes a pyrimidine derivative, contributing to its mechanism of action and pharmacological properties.
