Description:

ML188 is a covalent inhibitor of the mammalian target of rapamycin (mTOR). It was originally developed as an anti-cancer drug, but has also shown efficacy in treating autoimmune diseases and bowel disease. ML188 binds to the ATP-binding site of mTOR, which prevents the formation of complex 1, leading to its inhibition. This binding leads to a decrease in proinflammatory cytokines and chemokines such as TNFα, IL-6, and IL-12. The binding of ML188 is reversible, with dissociation constants in the low nanomolar range. The binding of ML188 leads to decreased expression levels for cyclin D1, c-Myc, p21 and p27 proteins. ML188 also inhibits growth factor activity by preventing phosphorylation of Akt/PI3K. In addition to inhibiting mTOR activity, ML188 may inhibit protein synthesis by interfering with ribosomal function.
