Description:N-((1R,6R)-6-Amino-2,2-difluorocyclohexyl)-5-methyl-4-(6-(1-methyl-1H-pyrazol-4-yl)pyrazolo[1,5-a]pyrimidin-3)yl)thiophene-2 -carboxamide (NCCXVII) is a potent and selective inhibitor of the human Btk protein. The IC50 values for NCCXVII were determined to be 1.0 μM in a cell free assay and 10 nM in an intact cell assay. It has been shown that NCCXVII inhibits ligand binding to the Btk receptor with a Ki of 0.8 nM, suggesting that it is a competitive inhibitor of ATP binding to the kinase domain of Btk.
