Hederagenin is a natural product that has been found to have cytotoxic effects on mouse tumor cells. It also inhibits the expression of inflammatory cytokines and chemokines, as well as the production of nitric oxide by activated macrophages. Hederagenin has shown anti-inflammatory activity in a variety of in vitro and animal models, including inhibition of prostaglandin synthesis. This agent also inhibits 3t3-l1 preadipocyte differentiation, which may be related to its apoptosis-inducing effects. Hederagenin binds directly to the response element on DNA and inhibits polymerase chain reaction (PCR) amplification of DNA. Hederagenin also alters mitochondrial membrane permeability, which may account for its significant cytotoxicity in HL-60 cells and 3T3-L1 preadipocytes.
