Fraxetin is a natural phenolic compound that has been shown to have antihyperglycemic effects. Fraxetin has a strong binding affinity for DNA and intercalates in the double helix, interrupting the progression of gene transcription. Fraxetin is also able to inhibit the growth of cancer cells by inducing apoptosis. Fraxetin has been shown to induce cytotoxicity in human polymorphonuclear leukocytes (PMNL) and squamous cell carcinoma cells, as well as inhibit the expression of Bcl-2 protein in PC12 cells. This compound also interacts with cell membranes and alters their permeability, which may be responsible for its inhibition of polymerase chain reaction (PCR). The mechanism of action for fraxetin involves the response element found in polymer compositions. This response element is activated by nuclease-sensitive dsDNA sequences, leading to the production of antisense RNA transcripts that inhibit mRNA synthesis.
