Spadin is a quinazoline-based drug that was originally developed as an antidepressant. Spadin is a soluble guanylate cyclase activator, which increases levels of cyclic guanosine monophosphate (cGMP). The antidepressant response was demonstrated in patch-clamp experiments on mouse brain cells. Spadin also has been shown to increase levels of cGMP in the blood vessels and heart, and causes a decrease in systolic pressure in deficient mice. This effect may be due to the inhibition of phosphodiesterase by spadin or its analogues. Spadin has not been tested for its effects on depression in vivo, but it has been shown to have an antidepressant effect in vitro.
