GMC 1-161 is a small molecule that inhibits the ubiquitin ligase activity of the proteasome and plays a role in the regulation of cellular proliferation. GMC 1-161 has been shown to inhibit autophagy, an important pathway for maintaining homeostasis, and induce apoptosis in cancer cells by inducing phosphatase and protein kinase B/Akt pathways. GMC 1-161 also exhibits anti-angiogenic properties by inhibiting matrix metalloproteinases (MMPs) that are required for tumor development. GMC 1-161 has shown efficacy against CD1 mice, which are genetically predisposed to develop cancers at an early age. This drug may be useful in treating cancer stem cells because it inhibits PI3K/AKT pathways.
