2-[[2-[(2-Methylphenyl)methyl]-3-sulfanylpropanoyl]amino]-4-methylsulfanylbutanoic acid (thiorphan) is a thiourea and a potent vasodilator. It inhibits the synthesis of angiotensin II, a peptide that constricts blood vessels and raises blood pressure. Thiorphan lowers blood pressure in both normotensive and hypertensive rats by inhibiting angiotensin II production. The drug also lowers plasma renin activity and natriuretic peptides. Thiorphan also blocks the action of metalloendopeptidase, an enzyme that breaks down atrial natriuretic peptide, leading to increased levels of this hormone in the bloodstream.
