EPZ020411 is a potent and selective inhibitor of the enzyme dihydrofolate reductase (DHFR) which blocks the synthesis of tetrahydrofolate, an essential cofactor for DNA synthesis. EPZ020411 has been shown to inhibit tumor cell growth in cultures and in organoids derived from patient tumors. It has also been shown to be active against a range of bacterial and fungal pathogens. EPZ020411 inhibits DHFR with an IC50 value of 0.6 µM and does not inhibit other enzymes such as thymidylate synthase or methionine synthase.
