PHA 767491 is a small molecule inhibitor of the kinase enzyme CDK8. It is used in vitro to study the effects of CDK8 on transcriptional regulation. PHA 767491 has been shown to inhibit the growth of cancer cells by inhibiting the activation of MCL-1, which regulates mitochondrial membrane potential and apoptosis. PHA 767491 also blocks cell proliferation and induces cell death in mouse hepatocyte-like cells. This drug may also have therapeutic potential for autoimmune diseases, as it has been shown to inhibit TNFα production by inhibiting nuclear factor kappa B (NF-κB) activation.
