GN 44028 is a proton pump inhibitor that has been shown to inhibit the production of tumor cells in vivo. It has been found to be effective against various cancers, including breast cancer, colon cancer, prostate cancer, and leukemia. GN 44028 inhibits acidic glycolysis by binding to the enzyme carbonic anhydrase II (CA-II). CA-II is involved in the conversion of carbon dioxide and water into bicarbonate and protons. When GN 44028 binds to CA-II it blocks the conversion of carbon dioxide into protons and prevents acidic glycolysis. This leads to a decrease in pH within the tumor environment and an increase in pH outside of the tumor environment. The increased pH outside of the tumor environment allows for more oxygen molecules to bind with hemoglobin, which can then be transported into the tumor microenvironment. This increases oxygen levels in this area, which can lead to cell death through oxidative damage or apoptosis induction.
