Description:

SIS3 is a small molecule that inhibits the TGF-β signaling pathway. It has minimal toxicity in preclinical models and is being evaluated as a potential treatment for eye disorders. SIS3 has been shown to inhibit the growth of 3T3-L1 preadipocytes by suppressing the expression of epithelial mesenchymal transition (EMT) markers, such as tgf-β1. It also prevents kidney fibrosis in an experimental model. The mechanism of action of SIS3 may be due to its ability to block the polymerase chain reaction and cell nuclei translocation, resulting in decreased levels of smad3 protein phosphorylation. Chemical inhibitors used in these experiments include 2-aminopurine, 5-aza-2′deoxycytidine, 5-azacytidine, daunorubicin, doxorubicin and vincristine.
