It inhibits the cleavage of amyloid precursor protein (APP) and prevents the accumulation of β-amyloid peptide (Aβ) in the brain. JNJ-40418677 blocks Aβ production by selectively inhibiting γ-secretase activity with little or no effects on other proteases. This drug has been shown to be safe and well tolerated in early clinical trials for Alzheimer's disease.
