Gemcitabine is a cytotoxic drug that inhibits cell growth by binding to the ribonucleotide reductase enzyme, which prevents synthesis of DNA. It has minimal toxicity and can be used in a model system to study apoptosis. Gemcitabine has been shown to inhibit the production of tumor necrosis factor-α (TNF-α) and serum enzymes, such as dismutase activity, in mice with lung cancer. This drug also has a toxic effect on respiratory system cells, which may be due to its ability to induce apoptosis. Gemcitabine is an inhibitor of the chk1 kinase and has been shown to inhibit polymerase chain reaction (PCR). This drug also binds to α1-acid glycoprotein and causes damage that leads to polymerization.
