ASP3026 is a potent anti-tumor agent that binds to the epidermal growth factor receptor (EGFR) and blocks the activation of downstream signaling pathways. ASP3026 has shown potent antitumor activity in vivo against human cancer cell lines, with selectivity against alveolar type cells over normal lung cells. It also has been shown to inhibit EGFR-dependent phosphorylation of ERK1/2, Akt, and PKCδ in vitro. The ASP3026 molecule contains two different sequences: an amino acid sequence that binds specifically to EGFR and a hydrophobic sequence for membrane penetration. The kinase selectivity of ASP3026 may be due to its ability to bind more tightly to EGFR than other kinases such as protein kinase C (PKC).
