HOE 32020 is a peptide that binds with high affinity to the extracellular domain of the human N-methyl-D-aspartate receptor (NMDAR) and blocks its activity. HOE 32020 is a potent inhibitor of glutamate binding to the NMDA receptor. The IC50 value for HOE 32020 is 1 nM. It has been shown to be an effective blocker of glutamate-induced cell death in rat primary cortical neurons and mouse cortical cells, as well as in hippocampal slices from rats and gerbils. HOE 32020 has also been shown to inhibit NMDA-induced intracellular Ca2+ release in rat hippocampal slices and inhibit NMDA induced excitotoxicity in rat primary cortical neurons.
