DO34 analog is a peptide that has been found to be an activator of the voltage-gated sodium channel. It was discovered by screening for ligands that bind to the extracellular domain of the Nav1.8 receptor. DO34 analog binds strongly to the extracellular domain of Nav1.8 and activates this receptor by triggering its allosteric site, which leads to an increase in the open probability of the channel. This leads to a significant increase in excitability of neurons and muscle cells as well as other effects on physiological processes such as blood pressure and heart rate.
