Diflapolin is an activating, epoxyeicosatrienoic acid (EET) analogue that is metabolized to soluble epoxide and activated by the enzyme epoxide hydrolase. Diflapolin has been shown to inhibit neutrophil migration in vitro, which may be due to its ability to inhibit production of soluble epoxides. Diflapolin also inhibits the activation of leukocytes and induces a decrease in the production of pro-inflammatory molecules such as prostaglandins. The molecular modeling studies show that diflapolin binds to the active site of soluble epoxide hydrolase and blocks access by EETs. This leads to a decrease in the levels of soluble epoxides, which may be responsible for diflapolin's anti-inflammatory effects.
