GNE-495 is a potent and selective small molecule activator of the TRPV1 ion channel. It is a member of the class of ligands known as capsaicinoids, which are often used as natural food additives to provide spicy or hot taste sensations. TRPV1 channels are expressed in sensory neurons and play an important role in pain sensation. GNE-495 selectively activates these channels and may have potential for treating conditions such as chronic pain and inflammation. GNE-495 has been shown to be a potent inhibitor of protein interactions with human recombinant TRPV1 receptors, with IC50 values of 0.5 μM for receptor activation and 1 μM for receptor inhibition.
TRPV1 receptors can be inhibited by excess extracellular calcium ions (Ca2+) or intracellular calcium ions (Ca2+). GNE-495 functions by binding to the intracellular site on TRPV1 receptors, thereby inhibiting Ca2+ influx
